An anime should have a bit of everything and emphasis on the one main thing that makes it great. Each anime/manga has its own uniqueness whether it be ecchi, comedy, or romance. But uniqueness and a bit of a side-dish as in everything helps a lot.
Character involvement and developement between the major/minors characters are the single most important thing in an anime/managa(my opinion of course)
The plot has to be synergized with the story so it makes sense. And location really doesn't matter. But it's the 4 main things that matter:
Characters
Plot
Story
Drawing Style(got to be consistant or otherwise readers might wonder why they change it now and then)
